Drug Discovery, Development & Delivery The Virtual Elaboration of Fragment Ideas: Growing, Merging and Linking Fragments with Realistic Chemistry

Fragment-based drug discovery seeks to identify relatively simple and low molecular weight molecules which interact with protein targets, then grow or link them into active leads. Molecular design software can be used to create realistic chemistry between and around fragments. Two key factors for success are: using computational techniques that are suited to handling small molecule fragments; a...

Aromatase inhibitors and arthralgia: a growing pain?

In the current issue of ONCOLOGY, Henry et al comprehensively review the current state of knowledge regarding the incidence and pathogenesis of aromatase inhibitor (AI)-associated arthralgia, and its potential implications in terms of delivering adequate, effective adjuvant therapy. It has clearly been demonstrated in several large adjuvant trials that the addition or substitution of AIs provid...

A fragment merging approach towards the development of small molecule inhibitors of Mycobacterium tuberculosis EthR for use as ethionamide boosters.

With the ever-increasing instances of resistance to frontline TB drugs there is the need to develop novel strategies to fight the worldwide TB epidemic. Boosting the effect of the existing second-line antibiotic ethionamide by inhibiting the mycobacterial transcriptional repressor protein EthR is an attractive therapeutic strategy. Herein we report the use of a fragment based drug discovery app...

Overcoming the Limitations of Fragment Merging: Rescuing a Strained Merged Fragment Series Targeting Mycobacterium tuberculosis CYP121

The fragment-based approach is a firmly established paradigm for developing small-molecule ligands as chemical tools and leads for drug development. At its heart, this powerful methodology involves the structure-guided design and synthesis of potent ligands from weak binding low-molecular-weight fragment molecules (typically <250 Da). There are three main strategies for the elaboration of initi...

Fragment-Based Identification of Influenza Endonuclease Inhibitors

The influenza virus is responsible for millions of cases of severe illness annually. Yearly variance in the effectiveness of vaccination, coupled with emerging drug resistance, necessitates the development of new drugs to treat influenza infections. One attractive target is the RNA-dependent RNA polymerase PA subunit. Herein we report the development of inhibitors of influenza PA endonuclease d...
